FOXO4-DRI is a D-retro-inverso peptide engineered to competitively disrupt the interaction between the FOXO4 forkhead-family protein and the p53 transactivation domain — an interaction senescent cells rely on to sequester p53 and evade apoptosis. In the foundational 2017 mouse study, researchers examined senescence, hepatic and renal function, and fur-density markers in naturally aged and doxorubicin-exposed (chemotoxicity) mouse models following systemic administration, reporting apoptosis selective to p16INK4a-positive senescent cells. Subsequent published work examined FOXO4-DRI's association with endothelial-cell and Leydig-cell senescence markers via the p53 signaling pathway in aged mice, and with senescent-cell clearance in expanded human chondrocyte cultures. FOXO4-DRI has not completed human clinical trials and holds no regulatory approval for any use.
