Semax is a synthetic heptapeptide (Met-Glu-His-Phe-Pro-Gly-Pro), an analog of the adrenocorticotropin fragment ACTH(4-10) with a C-terminal Pro-Gly-Pro extension. In rat studies, a single application was reported to increase hippocampal BDNF protein and trkB tyrosine phosphorylation, alongside multi-fold increases in BDNF and trkB mRNA, with researchers examining Semax's association with modulation of the hippocampal BDNF/trkB signaling system. Related rodent work reported parallel changes in NGF and BDNF gene expression across hippocampus, frontal cortex, and retina, and a rat model of cerebral ischemia-reperfusion examined genome-wide transcriptional changes — including suppression of inflammatory gene expression — associated with Semax administration. Semax has not completed human clinical trials and holds no regulatory approval for any use.
